Abstract:A PPDK gene named Si PPDK1 was identified in foxtail millet genome by sequence alignment.To reveal the response of Si PPDK1 to abiotic stress,the gene structure,protein characteristics,function,cis-elements,subcellular localization predictions and evolution of Si PPDK1 were analyzed and predicted by online tools and software.The quantitative real-time PCR(q RT-PCR)was used to detect the induced expression of Si PPDK1 under different stresses during seedling stage and under drought during key growth stages.The results showed that Si PPDK1 was located on chromosome 9 in the foxtail millet genome,containing 17 introns and 2 alternative transcripts.Functional domain analysis and multiple sequence alignment revealed that the Si PPDK1 protein has a very conserved sequence structure and is very similar to other plant PPDK proteins.q RT-PCR analysis showed that the expression of Si PPDK1 was strongly up-regulated under PEG,ABA,Na Cl and low temperature at its seedling stage.Further studies indicated that Si PPDK1 was involved in drought stress and different light intensity in jointing,heading and filling stage.It is speculated that Si PPDK1 is involved in abiotic stress,especially plays an important role under drought and light stresses at the heading stage.
