摘    要:单核苷酸多态性(single nucleotide polymorphism,SNP)广泛分布在植物基因组中,位点丰富,易于实现高通量分析。竞争性等位基因特异性PCR(kompetitive allele specific PCR,KASP)是一种对特定位点处的SNP以及插入和缺失(insertion-deletion,In Dels)进行双等位基因分型的技术,具有稳定性好、准确性高、无需合成特异荧光探针、检测成本较低和高通量等优点,近些年在医学和农学领域都有广泛的应用。本研究介绍了KASP技术的原理,综述了该技术在小麦等植物抗病遗传育种中的应用,并讨论了该技术的优缺点。

Abstract:Single nucleotide polymorphisms(SNPs)are widely distributed at abundant loci in plant genome,therefore are well-suited for high-throughput analysis.Kompetitive Allele Specific PCR(KASP)is a biallelic genotyping technology to detect SNP,insertion and deletion(In Dels)at a specific site.KASP technology has advantages over many other SNP genotyping technologies,because of its good stability,high accuracy,high throughput,free of specific fluorescent probes and low cost.In recent years,KASP technology has a wide range of applications,especially in the fields of medical and agricultural science.In this paper,the principle of KASP is introduced;application of KASP in breeding for disease resistance in wheat and other plants are reviewed.Moreover,the advantages and disadvantages of KASP technology are discussed.
