透明质酸对牛卵母细胞体外成熟影响的研究

摘    要:在改良的基础培养液中添加HA,主要探讨HA对牛卵母细胞成熟的影响。结果表明,当mTCM-199中HA浓度为3.0 mg/mL时,第一极体排出率和卵裂率显著高于其他各组 (P<0.05),初步表明3.0 mg/mL是无血清培养中HA的最佳浓度。当在mTC-199中添加 HA、BSA和OCS时,OCS组的第一极体排出率显著高于其他两组(P<0.05),分别为 83.40%、84.33%和92.17%,但HA组与BSA差异不显著(P>0.05);卵裂率三者差异均不显著(P>0.05);表明在无血清培养中HA可代替BSA,但是培养的效果不如血清。HA在 mTCM-199和mSOF基础培养基中,对卵母细胞的第一极体排出率和卵裂率无明显差异(P >0.05)。表明在无血清培养下,HA在复杂和简单的培养液中都支持卵母细胞的成熟。

Abstract:In this study, hyaluronic acid (HA) was added to the modified foundation medium, through which to mainly determine the effect of HA on in vitro maturation (IVM) of bovine oocytes. The result show that, the group with 3 mg/mL HA was significant higher than other groups in the frequency of first polar body extrusion (PB) and cleavage rate of oocytes(P<0. 05), this primitively expresses that 3 mg/ mL was the best concentration for free serum culture with HA. When we added HA, bovine serum albumin (BSA)or Oestrus Calf Serum(OCS)to TCM-199, the group with the OCS was significantly higher than other two groups (83.40%, 84.33% and 92. 17%, P<0. 05) in PB, but cleavage rate was no significant different in three groups P>0. 05; there were no different in PB of BSA and HA (P>0. 05). There for HA was a substitute for BSA in scrum-free medium to mature the oocytes, but the effect was not equal to blood serum in result of the culture, so HA can not fully replace the serum until it was further research. HA in TCM-199 and Synthetic Oviduct Fluid (SOF) have no effect on in vitro maturation of bovine oocytes in PB of oocytes and cleavage rate; it demonstrated that HA can favor the maturity of oocytes in complicated or simple free-serum medium.
