石斛兰的组织培养

摘    要:目的]研究适合石斛兰叶片愈伤组织的诱导、茎段腋芽的诱导、根系的诱导、增殖的培养基、生长调节剂的种类及浓度。方法]设置5种叶片愈伤组织诱导培养基,3种茎段腋芽诱导培养基,3种根系诱导培养基,在温度为25℃,光照12h,光照强度2000lx进行培养。结果]6-BA对石斛兰叶片愈伤的诱导有一定的抑制作用,较高浓度的2,4-D有利于叶片愈伤组织的诱导;MS+6-BA4.0mg/L+NAA1.0mg/L是茎段腋芽诱导最理想的培养基;添加2.0mg/L肌醇和10%的香蕉汁均能促进根系的诱导,后者的效果更明显。结论]以容易获得的石斛兰幼嫩茎段为外植体材料研究了石斛兰的组织快繁,为石斛兰的快速繁殖提供了可行途径。

Abstract:Objective] The study aimed to research the induction callus, induction axillary bud, induction root system, value-added medium and the kind and concentrations of plant growth regulators of the medium of Dendrobium orchid TS1. Method] Five kinds of mediums to initiate the blade callus, three kinds of mediums to initiate axillary bud and three kinds of mediums to initiate root system were designed, all of materials were cultivated under the conditions of 25 ℃, 12/12(day/night) and 2 000 lx. Result] 6-BA showed some inhibiting effects to initiate the blade callus, while higher concentrations of 2,4-D promoting blade callus initiation. MS+6-BA 4.0 mg/L +NAA 1.0 mg/L was the best medium to initiate the axillary bud. Both 2.0 mg/L inositol and 10% banana juice promoted root system initiation, banana juice was better. Conclusion] With easily gained stem section of Dendrobium orchid as tested materials to study its rapid propagation technique, the research pointed feasible route to rapid propagations of Dendrobium orchid.
