小麦条锈病菌菌种的纯化与保存方法

摘    要:通过对田间收集的小麦条锈菌进行分离和鉴定,建立了单孢子堆分离方法,并比较了不同储藏温度下保存不同时间后条锈菌对小麦的致病率,以探索条锈菌的保存条件。结果表明,单孢子堆分离法获得的菌系可满足后续研究;冷冻保存后菌种的致病率均大于93%,明显高于冷藏保存后的致病率,年限越长,差异性越明显,表明冷藏保存只适宜菌种的短期保存,而冷冻保存适宜于长期保存。

Abstract:Uredospores of Puccinia striiformis f.sp.tritici from fields were purified and identified to set up single pustule isolation method,which satisfied requirements for further study.The storage conditions were also explored by comparing their pathogenicity after stored at different temperatures for different years.The results showed that the pathogenicity rate of spores stored under frozen conditions was over 93%,much higher than that stored under cold storage condition,the difference more significant with longer years,which indicated that cold storage condition was fit for short-time saving of Puccinia striiformis while frozen condition was more proper for its long-time storage.
