丁酸梭菌早期干预对幼龄番鸭生长性能、肠道黏膜形态及肠屏障功能的影响

摘    要:本试验旨在研究丁酸梭菌早期干预后对幼龄番鸭生长性能、肠道黏膜形态及肠屏障功能的影响。选取1日龄、平均体重(47.3±3.9)g的番鸭160只,随机分对照组和丁酸梭菌组,每组8个重复。丁酸梭菌组灌服1 mL丁酸梭菌菌液(≥2×109CFU/mL),对照组灌服1mL生理盐水,连续灌服3d。分别于第7、10和14日龄采集回肠及盲肠组织,用于肠道形态及Claudin1、Claudin2、ZO-1和ZO-2紧密连接蛋白基因表达分析。结果显示:与对照组相比,丁酸梭菌组番鸭体重在第7和10日龄呈升高的趋势(P>0.05);第7和10日龄丁酸梭菌组番鸭回肠绒毛高度和绒毛高度与隐窝深度的比值均显著增加,而第7日龄隐窝深度显著降低;与对照组相比,第7日龄丁酸梭菌组Claudin1基因相对表达水平上调111%、Claudin2上调111%、ZO-1上调132%、ZO-2上调143%;第10日龄Claudin1基因相对表达水平上调111%、Claudin2上调243%、ZO-2上调108%;而盲肠的紧密连接蛋在第7日龄丁酸梭菌组Claudin2基因相对表达水平与对照组相比下调65%;第10和14日龄Claudin2分别上调46%和131%。在本试验条件下,丁酸梭菌早期干预可以在一定程度上提高幼龄番鸭的体重,有效改善番鸭肠道黏膜形态结构和提高肠道紧密连接蛋白基因的表达量。

Abstract:The purpose of this experiment was to study the effects of early intervention of Clostridium butyricum on body weight,intestinal mucosal morphology and intestinal barrier function of young Muscovy duck.A total of 160 day-of-hatch Muscovy ducks with average weight(47.3±3.9)g were randomly divided into 2 groups of 8 replicates.The test group was that 1 mL Clostridium butyricum solution(2×109 CFU/mL)were taken orally as soon as possible after incubation in the experimental group.Ducks in the control group received the same amount of saline.The medicine was discontinued for 3 days of continuous inoculation.The ileum and cecum tissues were collected at 7,10 and 14 days respectively for intestinal structure analysis and RT-PCR analysis of the relative expressions of Claudin1,Claudin2 ZO-1 and ZO-2,respectively.The results showed that compared with the control group,the body weight of Muscovite ducks increased on the 7 th and 10 th days in the experimental group.The villi height and the ratio of villi height to crypts depth increased significantly in the 7 th and 10 th day experimental groups,and the depth of the 7 th day recess significantly decreased.Compared with the control group,relative expression level of Claudin1 gene was increased by 111.22%,Claudin2 by 111.23%,ZO-1 by 131.72%,and ZO-2 by 142.92%in the 7 th-day old experimental group.Relative expression of Claudin1 gene was increased by 110.92%,Claudin2 by 242.80%and ZO-2 by 107.96%at 10 th day.Compared with the control group,relative expression level of Claudin2 gene was downregulated by 65.17%in the experimental at 7 th age,and Claudin2 was up-regulated by 46.48%and 130.54%at 10 th and 14 th ages,respectively.In this experiment,the early intervention of clostridium butyrate can improve the weight of young Muscovy duck to some extent.It can effectively improve the intestinal mucosal morphology and structure and increase the expression of intestinal tight-junction protein gene.
