平菇816菌丝生长过程中纤维素酶活性的变化特征

摘    要:以平菇816为研究对象测定了菌丝体不同生长时期的每日生长量、总生长量、生长速率以及3种纤维素酶酶活性,结果表明平菇816菌丝体的再生能力和生长势很强第1天的日生长量最大第1天至第6天是快速生长期占总生长量的72.17%之后进入平稳生长期,菌丝体平均生长速度为9.88mm/d,外切β1,4葡聚糖酶活性随着生长天数增加而增加而内切β1,4葡聚糖酶活性、β1,4葡聚糖苷酶活性和纤维素酶总活性在第5天达高峰最高和最低酶活性分别相差4.38倍、3.65倍、2.24倍,外切β1,4葡聚糖酶活性与每日菌丝体生长量存在显著正相关(P〈0.05)回归方程为y=0.0209x+1.2545,早期纤维素活性高低影响菌丝体生长纤维素酶活性与菌丝体生长呈现正相关外切β1,4葡聚糖酶在纤维素分解过程中的起到关键作用显著影响菌丝体代谢能力进一步影响了菌丝体的生长势.

Abstract:The pres-ent study was set to determine daily growth quantity , total growth quantity and growth speed of mycelium growth capacity and 3 kinds of mycelium growth capacity by using PLeurotus ostreatus 816 as research object. The results indicat ed t ha t regener at ed capacit y of P Leurotus ostreatus 816 and growth irnpet urs were strong rt he daily growth of fourth day was the maxitn um vit s fast growth period occurred on the first day and sixth day-which occupied 72.47% of total growt h vt hen came to the st ealy period s aft er the sixth day the mycelium growth were in stable growth phase. The average growth speed was 9.88 mm/d.During growth days exo-β-1, 4 glucanase (CBH) activity was raised. How ever , endo-β-1, 4 gluca na se (EC) activity,β-1,4 glucosidase (BC) activity and total cellulase activity reached the highest peak in fifth day. There were 4.38-3.65-2.24 times between the highest and the lowest activity from EC-BC and total cellulase activity, respectively. There were positive relationship among cellulase activity and the rn ycelimn growth. There were the positive relationships bet ween the exo-β-1,4 glucanase activity and mycelium daily growth quantity ( P 〈0.05), and the regression equation was y =0.0209x + 1.2545. There were differences among these different cellulase activities and they affect the mycelium growth of PLeurotus ostreatus in the early stage. There were positive relationships bet ween cellulase activity and the mycelium growth. Exo-β-1,4 gluca nase activity plays an important role in metabolic and mycelium growth capacity.
