土壤磷酸酶测定中不同缓冲溶液对显色液吸收波长的影响

摘    要:确定了土壤磷酸酶测定时最大吸收波长以及显色时应加入的最佳pH缓冲溶液。研究结果表明,加入pH为8.0~10.0的缓冲液时显色时间快,在可见光区的最大吸收峰在604 nm处,其中以pH 9.0的吸光度最高,摩尔吸光系数为1.717×104L/(mol.cm),灵敏度较高。因此确定加入的最佳缓冲溶液pH为9.0,同时选择604 nm作为最大吸收波长。

Abstract:The maximum absorption wavelength and the optimal pH buffer solution in determination of soil phosphatase had been determined.The results showed that the buffer pH in the range of 8.0~10.0 colored fast,and the maximum absorption peak was 604 nm.The absorbance of the solution added the buffer(pH=9.0) was the maximum,and the molar absorptivity was 1.717 × 104 L/(mol·cm).The suggestion was that the optimal pH buffer solution was 9.0,and 604 nm was chosen in determination of soil phosphatase.
