羊驼酪氨酸酶基因部分cDNA序列的克隆及序列分析

摘    要:从3~4龄青年公羊驼背部皮肤中提取总RNA作为模板,采用RT-PCR技术扩增出TYR基因的部分Cdna序列,将其克隆进Pgem-T easy vector中,经限制酶酶切鉴定后进行测序,将测序结果与小鼠、人、猩猩等其它物种的TYR核苷酸序列进行比对,发现本研究获得部分序列为羊驼酪氨酸酶基因第三外显子,该序列比较保守.

Abstract:Total RNA extracted from the dorsal skin of 3-4 years alpaca was used to amplify partial cDNA encoding TYR with RT-PCR technique.The PCR product was cloned into pGEM-T easy vector,and the recombinant plasmid was digested with restriction endonuclease,and then the positive plasmid was sent to be sequenced.After being aligned with the related cDNA sequences of mouse,human,and chimpanzee,it was found the partial cDNA sequences was the third exon of Alpaca TYR and the sequence was conserved.
