摘    要:为寻找一种合适的提取球虫基因组DNA的方法,试验采用氯化苄法(BC)和常规提取DNA的方法提取了鸡柔嫩艾美耳球虫孢子化卵囊基因组DNA。氯化苄的化学性质类似于苯酚,在65℃条件下与蛋白质和几丁质等发生反应,能够有效地破坏球虫卵囊壁,可用于大规模提取基因组DNA;常规方法提取DNA的程序是用蛋白酶K消化处理,用酚/氯仿抽提,再用氯仿抽提,部分步骤需要重复。2种方法对比试验结果表明,二者均可用于鸡球虫基因组DNA的提取,所获得的DNA数量和质量都很高,可用于各种分子生物学实验。但BC法具有简便、快速、经济实用的特点,更适用于球虫卵囊大量样品的DNA快速提取。

Abstract:In order to look for a suitable method for extracting high quality genomic DNA from Eimeria tenella sporulated oocysts,the benzyl chloride method was used in comparision with the usual method.Benzyl chloride,which had chemical characteristics similar to phenol,could efficiently break sporulated oocyst wall.The results indicated that,the two methods were efficient for extracting genomic DNA.The quantity and quality of the DNA extracted by these two methods were all high.But the benzyl chloride method was not only simple and fast,but also inexpensive.Therefore,the benzyl chloride method is fit for large scale preparation of genomic DNA from E.tenella sporulated oocysts.
