大麻试管苗生根培养基的优化

摘    要:目的]寻找适合大麻试管苗生根的培养基。方法]以大麻试管苗为试材,设置不同的生长激素、基本培养基、蔗糖浓度和pH值处理,统计不同处理试管苗的生根数、腋芽数、茎粗和根长等指标,并计算根的诱导率。结果]0.1 mg/L IBA和0.05 mg/L NAA处理的试管苗平均生根数最多,达2.45条/株,而0.5 mg/L IBA和0.05 mg/L NAA处理的试管苗平均生根数最少。1/2 MS和MS基本培养中的试管苗平均生根数较多,分别达到2.05和2.45条/株。30 mg/L蔗糖处理的试管苗平均生根数和根诱导率均显著高于其他处理,分别为2.25条/株和80.0%;pH值为5.8的处理试管苗平均生根数和根诱导率均最大,分别达到2.35条/株和75.0%。结论]大麻试管苗生根的优化培养基为1/2 MS+0.1 mg/L IBA+0.05 mg/L NAA+30 mg/L蔗糖,培养基pH值为5.8。

Optimization of Rooting Medium for Tube Seedling of Cannabis sativa
Abstract:Objective] The study was to find the rooting medium suitable for tube seedling of Cannabis Sativa.Method] With tube seedling of C.sativa as the tested material,the treatments of different hormone combinations,basic medium,sucrose concn.and pH value were set up.The number of rooting,axillary bud number,stem thickness and root length of the tube seedling in different treatments were made for statistic and the induction rate of root was calculated.Result] The average rooting number of tube seedling in treatments with 0.1 mg/L IBA and 0.05 mg/L NAA were the most,reaching 2.45 trips/plant and that in treatments with 0.5 mg/L IBA and 0.05 mg/L NAA were the least,and that in treatments with 1/2 MS and MS basic medium were more,being 2.05,2.45 strips/plant resp.The average rooting number and the induction rate of root of tube seedling in treatments with 30 mg/L sucrose was significantly higher than that in other treatments,being 2.25 strips /plant and 80.0% resp.,and that in treatments with pH 5.8 was the biggest,being 2.35 strips /plant and 75.0% resp.Conclusion] The optimal rooting medium for tube seedling of C.sativa was 1/2 MS+0.1 mg/L IBA+0.05 mg/L NAA+30 mg/L sucrose with pH 5.8.
