山银花叶中绿原酸的提取分离纯化方法研究

摘    要:本文以山银花叶为试验材料,通过正交试验优选出绿原酸最佳提取方法;使用大孔树脂对提取液进行粗分离,优化大孔树脂型号、上样液pH值、树脂对绿原酸的饱和吸附量及树脂洗脱条件;在乙酸乙酯纯化工艺中,考察萃取时间、萃取剂用量、萃取料液浓度、萃取次数,得到最佳萃取工艺。结果表明,在所确定的最佳提取、分离、纯化条件下,绿原酸纯度可达到95%以上(HPLC)。该提取分离纯化方法对山银花叶中有效成分绿原酸的提取率高、稳定性好,生产成本低廉,适用于工业化生产,是一种较好的高含量绿原酸提取物制备工艺。

Study on Extraction, Separation and Purification of chlorogenic acids from the Leaves of Lonicera confusa
Abstract:Objective To study the extraction, separation and purification of chlorogenic acid from the leaves of Lonicera confusa, so that the purity of chlorogenic acid can reach more than 95% (HPLC). METHODS The optimum extraction method was selected by orthogonal test with chlorogenic acid as the evaluation index. Macroporous resin was used to roughly separate the extract and optimize the type of macroporous resin, the PH value of sample solution, the saturated adsorption capacity of resin to chlorogenic acid and the resin elution conditions. In the purification process of ethyl acetate, the extraction time, the amount of extractant, the concentration of extractant solution and the extraction times were investigated. To the best extraction process. Results Under the optimum conditions of extraction, separation and purification, the purity of chlorogenic acid could reach more than 95% (HPLC). CONCLUSION The extraction, separation and purification method has high extraction rate, good stability, low production cost, and is suitable for industrial production. It provides a better preparation process of chlorogenic acid extract with high content..
