摘    要:以成熟胚愈伤组织为转化受体 ,经农杆菌介导法将显性广谱白叶枯病抗性基因 Xa2 1导入高感白叶枯病的水稻广亲和恢复系 SWR2 0中 ,共获得 4 3个独立的转基因株系 ,1 0 0余株转基因水稻植株。经 GUS活性、潮霉素抗性检测及PCR分析表明 ,外源基因已整合到转基因水稻基因组中 ;白叶枯病原菌接种试验表明 ,大部分 T0 代转基因水稻植株的抗病性有明显提高 ,多数植株表现抗或高抗 ,个别为感病。经对农杆菌介导的水稻转化技术作进一步改进后 ,GUS瞬间表达率可高达 92 .5% ,稳定转化频率达到 2 7.6%

Abstract:A wide compatibility restore line SWR20 was efficiently transformed with Xa21 gene via Agrobacterium tumefaciens, using the calli derived from mature embryos as the recipients. A total of 43 independent transgenic plant lines, more then 100 transgenic plants were regenerated, and most of these transgenic plants could grow normally. The results of GUS expression, hygromycin resistance and PCR analysis showed that the foreign genes had been integrated into the genome of the transgenic rice. After inoculation with the HB12 race of Xanthomonas oryzae Pv. oryzae (from Northern of China), most of the T 0 transgenic plants were resistant or highly resistant to rice bacteria blight (BB), and only a few of transgenic plants showed sensitivity to the BB pathogen. Some factors influencing Agrobacterium mediated transformation of rice were also improved, the efficiency of GUS transient expression could reach 92.5% and the stably transformation efficiency was increased to 27.6%.
