Identification of the pathogenic virus on Ailanthus altissima

Abstract:Leaves from Ailanthus altissima showing severe symptoms were selected and indexed by antigen-coated plate (ACP)-ELISA. Tobacco mosaic virus (TMV) was found in all the samples tested. By using dsRNA as template, one-step RT-PCR was performed to confirm the presence of TMV with specific primers designed for the partial movement protein gene (MP), and a fragment of about 470 bp(GenBank GU167975)was amplified. Sequence analysis demonstrated that Ail isolate showed 75.5%~99.6% identity with other TMV isolates, and shared the highest identities of 99.6% at nucleotide (nt) level with isolate B8 from Spain. From the above, it can be concluded that the virus disease pathogen infecting Ailanthus altissima was TMV.
