摘    要:以一株产漆酶树舌灵芝的菌丝片段为试材,利用紫外线和硫酸二乙酯逐级诱变;采用愈创木酚显色圈初筛突变菌株,并通过摇瓶复筛对优势菌株产酶能力进行验证,以期选育稳定高产漆酶菌株。结果表明:最终诱变获得一株遗传稳定的高产突变株,与原始菌株相比,漆酶活性提高63.3%,同时发酵周期缩短1 d。

Abstract:The mycelium fragments of Ganoderma applanatum were used as test material,using ultraviolet ray and diethyl sulfate to induce mutagenesis step by step,in order to select and breed stable and high yield laccase strain,the mutants were primary screened in guaiacol-containing SPDA plates,and then the ability of the dominant strains to produce laccase was verified by triangular bottle fermentation experiment.The results showed that a mutant strain with high yield and genetically stable was obtained finally,laccase activity was 63.3%higher than that of the original strain,and the fermentation cycle was shortened by 1 day.
