柱杨的高效离体快繁技术

摘    要:[目的]为利用组织培养技术培育柱杨新品种提供理论依据。[方法]以柱杨试管苗为材料,取其茎段为外植体,筛选最佳芽诱导培养基;分别以试管苗的叶片、茎段和叶柄为外植体,研究不同组织的再生芽诱导率;配制不同激素浓度的生根培养基诱导组培苗生根,筛选最佳生根培养基。[结果]培养基MS+2.0 mg/L6-BA+0.40 mg/LIAA诱导再生芽的效率最高,平均每块外植体再生芽8.6个,但芽的质量较差。综合考虑,较理想的芽诱导培养基为MS+1.0 mg/L6-BA+0.50 mg/L IAA;柱杨茎段产生再生芽的能力最强,叶柄次之,叶片最差;培养基1/2MS+0.2 mg/LNAA与1/2 MS+0.1 mg/LNAA诱导组培苗的生根率最高,每株平均生根数达8条以上,但根的质量较差。综合考虑,最佳生根培养基为1/2 MS+0.2 mg/LIAA。[结论]柱杨最佳芽诱导培养基为MS+1.0 mg/L6-BA+0.50 mg/LIAA,最佳生根培养基为1/2 MS+0.2 mg/LIAA。
