摘    要:热激蛋白70(heat shock protein 70,HSP70)是广泛分布于生物体内的重要分子伴侣,在环境应激和热适应中起着重要作用。本研究对诗神袖蝶 Hsp70s 进行了全基因组鉴定,共获得12个 Hsp70s 基因,其编码蛋白分子量均在70 kDa 左右。进化分析表明,Hsp70s 在分子系统发生树中聚为2类,分别为应激诱导型(HSP70)和组成型(HSC70)。诗神袖蝶和果蝇组成型 Hsc70s基因存在1∶1的直系同源关系,而应激诱导型 Hsp70s 则同一物种聚为一枝,表明 Hsc70s 在诗神袖蝶和果蝇物种分化前业已发生基因扩增,而 Hsp70s 则是物种形成后发生世系特异扩增而产生的多个拷贝。我们对诗神袖蝶 Hsp70s 基因在化学感受器官中的表达进行了分析,除 HmelH-sp 68、HmelHsp 70A 和 HmelHsc 702在化学感受器官中不表达或表达量极低外,其他基因均有明显的表达信号(FPKM>1),而且在雌雄个体间具有相似的表达信号。这一研究有助于拓展我们对昆虫 Hsp70s 进化和功能的认识。

Genomic Characterization and Evolution of HSP70 in Heliconius melpomene
Abstract:Heat shock protein 70 (HSP70)is widely distributed in organisms,which plays impor-tant roles in environmental stresses and heat adaptation.In this study,Heliconius melpome-ne Hsp70s were identified in the whole genome,and 12 Hsp70s were characterized,whose molecular weights were all around 70 kDa.In the phylogenetic tree,heat shock protein 70 proteins were clustered into two group,including stress-induced Hsp70s and constitutive Hsc70s.Relatively,most of the Hsc70s in H .melpomene and Drosophila melanogaster showed 1 to 1 orthologous relationship,while stress-induced Hsp70s group clustered within each species,suggesting that Hsc70s were duplicated before speciation of H .melpomene and D .melanogaster.We analyzed the expressions of HmelHsp70s family in the chemosensory tissues of H .melpomene adults.Except for HmelH sp 68,HmelH sp 70A and HmelH sc 70 2,all the others were expressed in those tissues (FPKM>1).This study might help us un-derstand the evolution and function of Hsp70s in non-model species.
