摘    要:[目的]为了进行气单胞菌几丁质酶产酶条件的研究。[方法]从自然死亡家蚕中肠围食膜中分离的气单胞菌进行产酶优化试验。其优化条件为:粉状几丁质1.2%,K2HPO4 0.12%,乙酸铵0.02%,发酵72 h,温度为30℃,起始pH值为8.0,培养基装量为15%。[结果]结果表明:该菌株所产几丁质酶的最适反应温度为25℃,pH值5.5,耐温性较差,但pH值5.0~9.0的酶活力较稳定(>88%)。[结论]该研究提供了提高几丁质酶产量的途径,但对该菌株进行基因改造或诱变育种,充分发掘其产酶潜力,对其几丁质酶基因进行克隆、表达等仍需进一步研究。

Abstract:[Objective] The aim of this paper was to study the condition for chitinase produced from Aeromonas sp.[Method] Aeromonas sp.was isolated from Bombyx mori.And its condition to produce chitinase was studied,which were 1.2 % powder chitin,0.12 % K2HPO4,0.02 % ammonium acetate,8.0 initial pH,15 % culture medium volume,72 h culture time and inoculated in 30 ℃.[Result] Result showed that the optimum condition for Aeromonas sp.to produce chitinase were 25 ℃ and pH 5.5.It was poor in heat resistance but was stable at pH 5.0~9.0 with at least 88 % activity.[Conclusion] Approaches to improve chitinase yield were provided in this paper.But there still needed further study in gene modification,mutation breeding,tapping the enzyme-producing potential,chitinase gene cloning and expression.
