猪流行性腹泻病毒SHpd/2012株Nsp8基因分析及真核表达

摘    要:应用生物信息工具和分子克隆技术对PEDV SHpd/2012株Nsp8基因编码的蛋白进行了结构和功能分析以及体外表达。结果显示,NSP8蛋白是一个含有194个氨基酸的亲水性蛋白,不含信号肽和跨膜区。SHpd/2012毒株高度保守的NSP8氨基酸序列与CV777、AJ1102等毒株同源性高。NSP8蛋白也是一个单体蛋白,共有5个优势抗原表位。此外,本研究成功地在293T细胞内表达了NSP8蛋白,为进一步研究猪流行性腹泻病毒NSP8蛋白的功能提供了理论基础。

Abstract:Bioinformatics tools were used to study the structure and function of NSP8 protein in strain SHpd/2012 of porcine epidemic diarrhea virus(PEDV).The results show that NSP8 is a hydrophilic protein,which contains 194 amino acids,without signal peptide and transmembrane region.Its conserved amino acid sequence is highly homologous to strains CV777 and AJ1102.Furthermore,the results also show that NSP8 is a monomeric protein with five dominant epitopes and can be successfully expressed in 293T cells,which provides a theoretical basis for further study of the function of PEDV NSP8.
