通过花粉管途径将抗虫基因(CpTI)导入小麦的研究

摘    要:利用带有CpTI基因的pAPCpTI-1-D质粒通过花粉管途径的不同方法导入冬小麦92(8)802,种子发芽经40mg/L卡那霉素液筛选,得到18株绿苗,移栽后成活10株。提取这10株小麦植株叶片的DNA,经PCR检测和PCR-Southern杂交两次鉴定分析,证明去柱头滴加法中的5株小麦CpTI基因已整合到其基因组中,获得5株转基因92(8)802小麦植株,长势良好,并得到饱满的种子。

Study on Introduction of Vermin-Resistance Gene(CpTI) into Wheat through Pollen-Tube Pathway Method
Abstract:The plasmid(pAPCpTI-1-D) with CpTI gene was introduced into winter wheat 92(8)802 through pollen-tube pathway,eighteen green seedlings were obtained after screening the progeny in 40mg/L Kanamycin solution,and ten of them survived after transplanting.Results of PCR and Southern hybridization analysis proved that five plants treated by dropping DNA liquid after cutting off the stigma had integrated the CpTI gene into their genome.These five putative transgenic plants grew well and had full seed.
