富有柿果ACC氧化酶cDNA的克隆及其序列分析

摘    要:以富有柿果为材料,用RNA提取试剂盒提取总RNA,根据已报道的柿果ACC氧化酶(1-aminocyclopropane-l-carboxylic acid oxidase,ACO)基因的序列设计并合成1对引物,通过RT-PCR方法获得1条约1.2 kb特异片段,把该片段连接到pGEM-T easy vector上进行测序,其全长共1 237 bp,编码区697 bp,共编码231个氨基酸残基。序列分析结果表明,该序列与DK-ACO1的cDNA序列同源性达99%,氨基酸的同源性达98%。

Abstract:Total RNA was extracted from Fuyu persimmon fruits and reverse transcribed to first strand of eDNA. A pair of primers were designed and synthesized based on the sequence reported. The specific PCR product of 1-aminocyclopropane-1-carboxylic acid oxidase was obtained using the first strand of eDNA as template. The product was ligased to pGEM^8 -T easy vector and sequenced. The sequencing data showed that the PCR product was 1 237 bp with 697 bp of coding region, encoding 231 predicted amino acid residues. Comparison with the eDNA sequence from DK-ACO1 indicated the homology was 99 %, and amino acid identities were 98 %.
