摘 要: | 为探索SLC27A1基因对牛肉质性状的影响,选取中国西门塔尔牛太行类群牛公牛、阉牛各20头为研究对象,采用PCR法分析SLC27A1的基因遗传多态性。检测到SNP位点为S5650:TC,该位点存在有3种基因型,其中CC基因型频率较高(62.5%),TT基因型频率最低(12.5%)。采用GLM对肉质相关性状与SLC27A1基因进行关联分析,结果表明,剪切力、肉色a、肉色b表型中,CC与TT及CT与TT差异均极显著(P0.01),其他性状间差异不显著(P0.05),阉牛与公牛间对肉质的弹性与咀嚼性差异显著(P0.05)。采用Real-time PCR分析,CC基因型在肌肉与脂肪中的表达量比基因型CT与TT差异显著(P0.05),阉牛与公牛之间的表达量在肌肉中差异极显著(P0.01)。SLC27A1基因S5650:TC位点突变对中国西门塔尔牛太行类群肉质性状存在一定的遗传效应。

Correlation Analysis on Polymorphism of SLC27A1 Gene and Its Relationships with Meat Quality Trait in Chinese Simmental Taihang Group |

Abstract: | 20 bulls and bullocks were chosen respectively as the research object in Chinese Simmental Taihang group to ex-plore SLC27A 1 genetic influence on beef quality traits. Genetic polymorphism of SLC27A 1 was analyzed by PCR. One T→C mutation was detected at 5650 bp of SLC27A 1 gene. There are three kinds of genotype in the loci, CC, TT and CT. And the CC genotype frequency was higher (62.5%), TT genotype frequency lowest (12.5%). The relationships between the geno-types and meat quality traits were analyzed by GLM. The results showed the difference was significant (P<0.01) between genotype CC and TT, CT and TT in the phenotypes of shear force, color a and color b. The difference in elasticity and the chewiness was significant (P<0.01) between bullocks and bulls. The Real-time PCR was used to study the expression of dif-ferent genotype in tissues. The results showed that the expression difference between CC genotype and CT and TT genotype was significant in muscle and fat for SLC27A1(P<0.05). And the difference was extremely significant between bullocks and bulls in the muscles (P<0.01). This study proves that S5650:T > C locus mutation of the SLC27A1 gene maybe have some ge-netic effect on meat trait in Chinese Simmental TaiHang group. |
