小麦种子醇溶蛋白聚丙烯酰胺凝胶电泳技术的简化研究

摘    要:在ISTA小麦和大麦种子醇溶蛋白聚丙烯酰胺凝胶电泳鉴定品种的标准程序的基础上,对影响电泳效果的各主要因素进行逐一筛选、改进,尤其是实验设计的NaOH—醋酸—甘氨酸缓冲系统和先低后高再低的电泳方式使此项技术具有快速、分辨率较高、重复性较好、易剥胶、操作简单、经济实用等优点,适于一般实验室利用小麦种子醇溶蛋白技术对小麦进行大量的快速简捷的筛选、鉴定等初步研究。

Abstract:Based on the standard procedure of polyacrylamide gel electrophotesis set by IST A for the analysis of the gliadin of wheat and barley seed for the varie ty identification,we screened the main influence factors which would affect the electrophoresis,and tried to improve them to get the best combination.Accordi ng to the results,we established an optimal procedure,it had the buffer system o f Sodium Hydroxide-Acetic Acid-Glycine,and the voltage of electrophoresis sho uld be lower at beginning,then higher for a while,and lower in the end.This m ethod had the properties of high resolution and stability,using less samples,a nd the gel can be easily desquamated than the other methods.Furthermore,this m ethod is easily to operate,more economical,and spend less time than the standa rd procedures,so it can be used in many laboratories to study the gliadin of wh eat.
