农杆菌介导北方粳稻转化体系的研究

摘    要:通过农杆菌介导的方法将Bar-Bt基因(抗除草剂基因和抗虫基因构建在同一载体上的双基因)整合到北方优质粳稻龙稻6号、松粳9号等品种中;分别以供试品种的幼胚和成熟胚为外植体诱导愈伤组织,携带有Bar-Bt基因双元载体PCAMBIA1301号的根癌农杆菌EHA105为载体,GUS基因的瞬时表达率为衡量指标,对影响农杆菌介导的龙稻6号、松粳9号等品种的转化效率的主要因素进行了研究。结果表明:农杆菌侵染愈伤组织的菌液浓度以D(600nm)=0.8~1.0较为适宜,最佳共培养时间为2~3 d,预培养时间为5 d,共培养培养基中添加100μmol·L^-1乙酰丁香酮的转化效率有明显提高。

Abstract:With juvenile embryo of grain-oryza of North of China as explant, Agrobacterium tumefaciens strain EHAl05 carrying Tiplasmid PCAMBIA1301 with Bar-Bt gene as vector,and the transient expression rate of GUS gene as measuring index,main factors that affected transformation efficiency of grain-oryza of North of China mediated by Agrobacterium tumefaciens were studied in this article. Results showed that the proper concentration of bacteria solution to infect callus was D(600nm)-0.8~1.0,the most suitable co-cultivation time was 2~3 days, pre-cultivation time was 5 days, and addition of 100μmol·L^-1 Acetosyringone improved transformation efficiency considerably.
