摘    要:以"珲春桃"与"正满早生"杂交获得的32株F_1代群体为材料,对影响桃AFLP技术分析体系的因素作了详细的探讨,优化出一套稳定、可靠的桃AFLP反应体系。经优化,得出最适宜的结果:酶切时间为6h、Mg- Cl_2浓度为1.75 mmol/L、预扩增产物稀释倍数为10倍。利用优化后的桃AFLP分析体系对该技术的重复性进行了验证,重复率可达90%。

Abstract:In this study,32 plants of F_1 progeny,obtained by crossing Hunchun peach and Zhengmanza- osheng,were used as materials to study the factors influencing AFLP analysis system in peach.A set of steady and credible system was optimized.Through optimization,the best results are proven to be:diges- tion time 6 hours,concentration of MgCl_2 1.75 mmol/L and dilution times of pre-amplification product 10 times.The experiment of testing the repetition of the technique was conducted using the optimized AFLP analysis system,and the frequency reached 90%
