猪圆环病毒2型吉林株的分离与鉴定

利用猪圆环病毒2型(PCV2)特异性引物,对吉林省某猪场发生的具有典型断奶仔猪多系统衰竭综合征(PMWS)症状病猪的淋巴结、脾脏组织进行PCR检测。将检测阳性的病料研磨、高心、过滤除菌,接种到无PCV污染的PK-15细胞进行病毒分离。将PK-15细胞增殖所得的病毒纯化后,在电镜下观察到直径约为17 nm的圆形病毒样粒子。PCV2间接免疫荧光试验,可在细胞浆中观察到特异性荧光颗粒。PCV2特异性PCR检测细胞培养物,获得阳性结果。由此说明,分离出的病毒为猪圆环病毒2型。

Isolation and Identification of Porcine Circovirus Type 2 Jilin Strain

Lymph nodes and spleen of a swine with typical syndrome of PMWS broken out in one porcine farm in Jilin province were detected by polymerase chain reaction(PCR) with a pair of primers specific to porcine circovirus type 2(PCV2) The PCV2 PCR positive samples were abraded,sedimented by centrifu- gation,filtered and inoculated to the PK-15 cell line,which were not contaminated by PCV1.The viral particles were purified and observed with electronic microscope.They were 17nm in diameter.In the cy- toplasm,the specific fluorescence particles were observed in the frozen section stained by indirect im- munofluorescence test specific to PCV2.And the specific nucleotide was amplified from the cell culture by the PCR,too.According to the above results,the isolated strain belongs to PCV2.
