橙皮素对铅致大鼠睾丸损伤的保护作用

摘    要:为研究橙皮素对铅致大鼠睾丸损伤的保护作用,将24只健康雄性SD大鼠随机分为4组(对照组、铅组、橙皮素组、铅+橙皮素组),各组每日分别灌服生理盐水(0.9%)、醋酸铅(500mg/L)、橙皮素(50mg/kg)、醋酸铅(500mg/L)+橙皮素(50mg/kg),连续8周后,处死大鼠并收集睾丸。检测大鼠睾丸组织中丙二醛(MDA)含量、还原型谷胱甘肽(GSH)含量、谷胱甘肽过氧化物酶(GPx)活性和过氧化氢酶(CAT)活性。结果表明,铅可显著提高MDA含量和GPx活性(P<0.05),增幅分别为62.74%和65.67%,显著降低GSH含量和CAT活性(P<0.05),降幅分别为33.08%和45.88%;橙皮素能显著降低铅处理组MDA含量和GPx活性(P<0.05),显著提高GSH含量和CAT活性(P<0.05)。说明铅可导致大鼠睾丸组织脂质过氧化损伤,橙皮素对铅导致的损伤具有一定保护作用。

Abstract:The present study was conducted to investigate the protective role of hesperetin against lead acetate-induced oxidative stress in rats.Twenty-four SD male rats were randomly divided into 4 groups,including the negative control group,the hesperetin-treated group,the lead-treated group,and the lead plus hesperetin-treated group.Eight weeks later,all the rats were killed for collection of the testes.The levels of MDA,GSH,GPx and CAT content in testis were measured and the results showed that MDA content and GPx activity in rat testis following the lead exposure significantly increased(P<0.05) while the content of GSH and CAT activity in testis significantly decreased(P<0.05).Hesperetin alone did not induce any significant changes in biochemical chemistry of testis tissues.Compared the hesperetin antagonistic groups to the poisoning groups,the levels of MDA and GPx in testis of rat were decreased significantly(P<0.05) while the content of GSH and the activity of CAT were increased significantly(P<0.05).The results suggested that hesperetin exhibited remarkable protect effects on lead-induced oxidative stress in the testis of rats.
