月月红中槲皮素·山奈酚的定性与定量研究

摘    要:[目的]通过对月月红中槲皮素、山奈酚的定性定量研究,拟建立其HPLC分析方法,为进一步研究民族民间药物月月红的药效学奠定基础。[方法]采用化学定性法确定月月红不同部位中的主要化学组分;色谱条件:Hypersil DJZ C18色谱柱(4.6 mm×200.0 mm,5μm);流动相:甲醇-0.4%磷酸(60∶40,V/V);检测波长:368 nm;流速:1.000 ml/min;柱温:30℃;理论塔板数按山奈酚和槲皮素峰计算不低于3 000,外标法计算。[结果]槲皮素在0.081 6~0.489 6μg线性关系良好(R2=0.999 6),平均回收率为101.6%,RSD为0.87%;山奈酚在0.069 6~0.417 6μg线性关系良好(R2=0.999 6),平均回收率为96.8%,RSD为1.01%。月月红叶中槲皮素为0.038 2%,茎为0.005 2%,全株为0.010 5%;月月红叶中山奈酚含量为0.027 6%,茎为0.002 1%,全株为0.009 6%。[结论]HPLC分析方法稳定、重复性较好;月月红不同部位均含有槲皮素、山奈酚,且叶含量高于全株和茎。

Abstract:[Objective] The qualitative and quantitative research on quercetin and kaempferol in Ardisia faberi Hemsl.were carried out to establish the HPLC analysis method,which provided a foundation for further study of pharmacodynamics of folk medicines-Ardisia faberi Hemsl..[Method] The main chemical compositions in different parts of Ardisia faberi Hemsl.were determined by using qualitative method.HPLC analysis was carried out based on Hypersil DJZ C18 chromatographic column(4.6 mm×200.0 mm,5 μm) and methanol-0.4% phosphoric acid(60∶40) as the mobile phase,the detection wavelength was 368 nm and the flow rate was 1.000 ml/min as well as the column temperature was 30 ℃.The number of theoretical plates was no less than 3 000 what calculated according to quercetin and kaempferol.[Results] The linear range of quercetin was among 0.081 6-0.489 6 μg(R2= 0.999 6),the average recovery was 101.6%,RSD=0.87%.The linear range of kaempferol was among 0.069 6-0.417 6 μg(R2=0.999 6),the average recovery was 96.8%,RSD=1.01%.The content of quercetin in the leafage was 0.038 2%,in the stem was 0.005 2%,in the whole plant was 0.010 5%.The content of kaempferol in the leafage was 0.027 6%,in the stem was 0.002 1%,in the whole plant was 0.009 6%.[Conclusion] HPLC analysis method was stable and the repeatability was better,quercetin and kaempferol consisted in different parts of plant.The content of the quercetin and kaempferol in the leafage was higher than the whole plant and the stem.
