摘    要:[目的]为鼠李糖乳杆菌LT22微生态制剂的开发和应用奠定基础。[方法]以海藻酸钠、壳聚糖和明胶为主要壁材,利用微胶囊包埋技术制备出了鼠李糖乳杆菌LT22微胶囊产品,并通过稳定性试验和急性毒性试验研究鼠李糖乳杆菌LT22微胶囊的稳定性及安全性。[结果]鼠李糖乳杆菌LT22双层微胶囊呈现近似球形的白色颗粒,均匀分散,囊球直径为200~500μm,是理想的微胶囊粒径。该微胶囊具有良好的耐受胃酸性和耐受肠溶性,在常温37℃下保存期较短,在低温下保存期较长,在70℃下处理1h以内及100℃下处理30s内,其活菌数量下降的较少,说明制备的微胶囊具有较强的耐受短时高温的性能,且对小鼠无毒副作用。[结论]该试验所研制的双层微胶囊大大提高了鼠李糖乳杆菌LT22的抵抗力。

Preparation on Microcapsule of Lactobacillus rhamnosus LT22 and its Stability and Toxicity Test

Abstract:[Objective] The study aimed to lay the foundation for the development and application of microbial ecological agent of Lactobacillus rhamnosus LT22.[Method] With the sodium alginate,chitosan and gelatin as the main wall materials,the microcapsule products of L.rhamnosus LT22 were prepared by using embedding technique of microcapsule and their stability and security were studied through the stability test and acute toxicity test.[Result] The double-microcapsule of L.rhamnosus LT22 was the near-spherical white granules in homodisperse,with a diameter of about 200-500 μm,which was the perfect granule diameter of microcapsule.The microcapsules were better on resistance to the gastric acid and solubility of intestinal juice.Their storage period was shorter at the normal temperature of 37 ℃ and longer at the low temperature.When they were treated for 1 h at 70 ℃ and for 30 s at 100 ℃,their active bacterium number was decreased less,showing that the prepared double-microcapsule had the characteristic of resistance to the ephemeral high temperature and had no toxicity on the rat.[Conclusion] The double-microcapsule prepared in the test could greatly increase the resistance to L.rhamnosus LT22.
