蚕丝蛋白制备方法的研究

摘    要:以废蚕茧丝为原料,将脱胶丝素通过盐解、酶解、碱解等方法,制备了溶液状、粉末状丝素蛋白。以沸水提取丝胶蛋白,采用二次水解、碱解等方法,制备了溶液状、粉末状丝胶蛋白。采用SDS-PAGE蛋白质凝胶电泳和红外光谱等检测分析了蚕丝蛋白的结构特征,探讨了制备方法对蚕丝蛋白结构和产率的影响。结果表明,不同方法制备的丝素蛋白、丝胶蛋白具有不同的分子量分布和不同的产率,其分子结构以无规卷曲为主。

Abstract:Various silk protein solution including fibroin and sericin with different molecular weight were prepared with the treatment of Cacl2,boiling distilled water,alkali protease or NaOH reagent.The silk protein powder was obtained through lyophilizing the various protein solutions.The molecular weight of silk protein was determined by SDS-PAGE technique and the secondary structure was characterized through fourier transform infrared spectroscopy.The yield ratio of the proteins was also tested and compared.The results showed that silk protein prepared with different condition have different molecular weight distribution and yield ratio.The secondary structures of all protein were mainly random coil.
