摘    要:番茄黄化曲叶病毒属于双生病毒,根据其外壳蛋白基因保守序列设计简并引物,对杭州和海宁地区番茄上疑似番茄黄化曲叶病毒进行PCR检测,获得产物克隆到pMD18-T载体中,经测序和序列分析,表明杭州和海宁地区发生的病毒是番茄黄化曲叶病毒,并且两地区病毒核苷酸序列同源性达97.5%。

Abstract:Tomato yellow leaf curl virus(TYLCV)is a geminivirus transmitted by the whitefly Bemisia tabaci.A pair of degenerate primers was designed according to the conserved sequence of coat protein genes of TYLCV.The tomato samples showing TYLCD-like symptoms were collected from Hangzhou and Haining and detected by PCR for virus presence.PCR products(about 361bp)were then cloned into the vector pMD18-T.Sequence analysis indicated that the nucleotide sequences of the amplified products from Hangzhou and Haining have 97.5% identity,and they share the highest sequence identity with TYLCV,indicating these samples were infected by TYLCV.
