镉对体外培养鸡脾淋巴细胞膜蛋白组分影响的研究

摘    要:以分组对比的方法(加镉组、对照组)研究镉对体外培养的鸡脾淋巴细胞膜蛋白组分的影响。通过体外分离培养鸡脾淋巴细胞,加镉组在培养体系中加入终度为10μmoL/L氯化镉,而对照组加入同体积10.0mmoL/L的PBS,分别培养12、24、36、48和72h后收集细胞,提取膜蛋白,用SDS-PAGE的方法对膜蛋白组分进行分析。结果表明:区带Ⅲ、Ⅳ、Ⅶ蛋白的百分含量变化不明显;区带Ⅰ和区带Ⅷ蛋白的百分含量呈增加趋势,区带Ⅱ、区带Ⅴ和区带Ⅸ蛋白的百分含量降低,而且在48和72h加镉组的电泳图前沿出现了高分子聚合物,分子量为70900.00。可以得出镉对体外培养的鸡脾淋巴细胞的膜蛋白组分产生损伤的结论。

Abstract:This study was focused on influence of cadmium on chicken spleen lymphocytes memberane proteins components cultivated in vitro by the use of group comparison method as follows adding cadmium and control group. By means of isolation and culturing chicken spleen lymphocytes,cadmium group with adding the final concentration of 10 μmoL/L CdCl2 in the cell culture system and control group with adding the same volum liquid of 10.0 mmoL/L PBS after culture in 12,24,36,48 and 72 h by collection cells and extraction membrane protein with using the methods of SDS-PAGE to ana-lyze membrane protein group. The results showed that the percents of protein of Ⅲ,Ⅳ,Ⅶ zone had no significant change and Ⅰ,Ⅷ zone of those with a trend of increasing,but Ⅱ,Ⅴ,Ⅸ zone of those with a trend of decreasing,moreover in the 48 and 72 h adding cadmium group emerged in the forefront of electrophoresis polymers,the molecular weight of 70 900.00. Cadmium can be derived in vitro of chicken splenic lymphocyte membrane protein component of the conclusions arising from injury.
