| 摘 要： | 为探明从河南安阳棉区分离的棉花黄萎病菌的系统进化关系,利用真菌内转录间隔区通用引物ITS1和ITS4对来自安阳地区的棉花黄萎病菌内转录间隔区(ITS)进行PCR扩增,得到542bp大小的片段,经过克隆、测序和在GenBank中进行BLASTN分析,证明了该片段来自大丽轮枝菌的ITS区,也进一步证明该棉花黄萎病菌是大丽轮枝菌。同时,对该序列在NCBI的GenBank进行了登记,登记号为EU835817。利用该ITS序列与GenBank中搜索到的黄萎病菌ITS序列一起构建了相应黄萎病菌的系统进化树,在进化树上所搜索到的13个大丽轮枝菌都聚类到一个进化枝上,而且中国报道的植物黄萎病菌,包括安阳地区黄萎病菌在内的3个大丽轮枝菌在进化树上处于相邻的位置,表明:这3个菌可能具有相同的起源。这些结果有助于理解棉花黄萎病安阳菌株的进化地位,该ITS序列还可以应用于棉花黄萎病安阳菌株的特异分子鉴定。
