锦葵脉明病毒中国蜀葵分离物cp基因序列分析

摘    要:本研究利用双链RNA(double-stranded RNA,dsRNA)和非序列依赖性PCR扩增技术(sequence-independent amplification,SIA)对表现出条斑症状的感病蜀葵进行了病原鉴定,结果发现侵染蜀葵的病原为锦葵脉明病毒(Malva vein clearing virus,MVCV),这是锦葵脉明病毒在国内首次报道。为进一步明确锦葵脉明病毒中国蜀葵分离物(MVCV-AR)的来源及进化关系,克隆获得其外壳蛋白(coat protein,cp)基因(Gen Bank登录号为KX619649)。通过与其他MVCV分离物的核苷酸及氨基酸序列比对分析可知,MVCV-AR与Mexico的tomato分离物(FJ561293)的核苷酸同源性最高为90.5%;与来自Italy的DS-Ba-01分离物(FM 212972)的氨基酸同源性最高为98.3%。系统进化分析表明,MVCV-AR与DS-Ba-01分离物聚为一簇,形成独立的分支。

Abstract:To identify the potential viruses that may cause Althaea rosea streak disease, the genomic dsRNA was extracted, followed by sequence-independent amplification (SIA). Results showed that Malva vein clearing virus (MVCV) was the pathogen causing the symptom in the diseased plant. This is the first report of Malva vein clearing virus naturally occurring in China. To further characterize this isolate, the cp gene of MVCV-AR were cloned and sequenced (GenBank accession number: KX619649). Sequence alignment between MVCV-AR and other MVCV isolates showed that MVCV-AR shares the highest identities of 90.5% with tomato isolates (FJ561293) from Mexico at the nucleotide level, and 98.3% identity with DS-Ba-01 isolate (FM212972) from Italy at the amino acid level. Phylogenetic analysis suggested that MVCV-AR and DS-Ba-01 clustered together and formed an independent branch.
