2016年北京地区玉蜀黍黑粉菌遗传多样性分析

摘    要: 2016年从北京市10个区采集317份玉米瘤黑粉病样品,选择核糖体RNA内转录间隔区的ITS rDNA和28S rDNA基因作为持家基因,通过单核苷酸多态性技术(Single Nucleotide Polymorphism,SNP)分析北京市10个区玉蜀黍黑粉菌的遗传多样性。ITS rDNA基因分析表明,有3个菌株的序列存在1个颠换突变位点,产生两种单倍型,采用邻接法构建的系统发育树将317个菌株分为2个群体;28S rDNA基因分析表明,有54个菌株的序列存在1个颠换突变位点,产生两种单倍型,采用邻接法构建的系统发育树将317个菌株分为2个群体。中性检测表明北京市10个区的玉蜀黍黑粉菌符合中性进化模型。试验结果表明北京市10个区的玉蜀黍黑粉菌遗传多样性不高,遗传分化不明显。

Analysis of genetic diversity of Ustilago maydis from Beijing in 2016

Abstract:We collected 317 Ustilago maydis samples from 10 suburban districts in Beijing, and used ITS rDNA and 28S rDNA gene of internal transcribed spacer in ribosome RNA as housekeeping gene to analyze their gene-tic diversity through SNP. The analysis of ITS rDNA indicated that 1 transversion locus was found in 3 strains and detected 2 haplotypes, Neighbor joining(NJ) tree divided the 317 strains into 2 clusters; The analysis of 28S rDNA indicated that 1 transversion locus was found in 54 strains and detected 2 haplotypes, NJ tree divided the 317 strains into 2 clusters. Neutrality test showed that the evolution of U. maydis was according with neutral theory. The genetic diversity of U. maydis from suburban districts in Beijing was not high, genetic difference was insignificant.
