摘    要:以耐热菌为研究对象.比较了CTAB法、CTAB改进法、经典酶法、氯化苄法、SDS法等5种楼板制备方法.提取DNA直接电泳结果和PCR结果表明.经典酶法和氯化苄法均能提供较好质与量的DNA模板;综合考虑时间等因素选择氯化苄法为此5种模板制备方法中的最优。

Abstract:Five different DNA extraction methods,sueb as the CTAB,the improved CTAB,the enzyme method,the benzyl chloride method and the SDS method,were evaluated for their effeot in preparing PCR templates from the Alicyclobucillus acidoterrestris.The results of DNA electrophoresis and PCR showed that both methods of benzyl chloride and enzyme could obtain DNA samples with sufficient quantity and superior quality,But considering the enyme method spending more time,the benzyl chloride method is more efficient
