| 摘 要： | 从家蚕品种大造和C108及其F1，F2的后部丝腺提取的基因组DNA通过的RAPD扩增，在477个随机引物中,85.9%的引物在家蚕基因组有稳定的扩增产物，总扩增片段5155条，每个引物扩增片段1-23条，平均为12.6条，多态性片段数共1496条，占两品种中总片段数的29.0%，经多次重复及反复验证发现，只要严格控制扩增反应条件及保证基因组DNA和试剂等不污染，则能检测到稳定的DNA多态性片段，从而保证其稳定性与重复性。

| Abstract: | The DNAs, taken from the hind silk gland of Silkworm C 108 , Dazao and from their first and second generations, were used for amplification by means of RAPD and it showed that 85.9% of the 477 random primers had stable products. The total number of amplified bands was about 5 155, with the bands of each amplifier ranging from 1 to 23, which averaged in 12.6. The polymorphic DNA fragments observed were 1 496, which was 29.0% of the total amplified products. After extensive repeat and experiments on large scale, it strongly suggested that only in a strict amplified condition and without any pollution in every step did the DNA polymorphism show repeatability and stability. |
