| 摘 要： | 从河北唐山分离到一株疑似猪繁殖与呼吸综合征病毒,接种Marc-145细胞,经过4代盲传后,出现细胞病变,经鉴定为PRRSV,命名为TS株。根据GenBank公布的PRRSV JXA1株ORF3基因的核苷酸序列,设计并合成一对特异性引物,用RT-PCR方法扩增完整ORF3基因,将扩增产物连接到pMD19-T载体并转化克隆菌,将阳性重组质粒PMD-GP3进行序列测定与分析。应用DNA Star软件,将测序结果与国内外已发表野毒株和疫苗株的ORF3基因进行序列比较,并绘制系统进化树。这为研制PRRSV新型疫苗及诊断试剂奠定了基础。

| Abstract: | One PRRS virus strain TS was isolated from pigs of Hebei Tangshan after inoculated to Marc-145 cell.The full-length of ORF3 gene of PRRSV strain TS was amplified by RT-PCR with a pair of specific primers designed according to the ORF3 sequence of PRRSV JXA1 strain,and then cloned into plasmid pMD19-T.The recombinant plasmid named as pMD-GP3 was sequenced and analyzed.The sequence was compared with that of other PRRSV strains in GenBank by using DNAStar software and the phylogenetic tree was drawn.It laid th... |
