7Li离子束注入小麦干种子诱发M1代变异的SSR分析

摘    要:为了探讨7Li离子束处理对植物种子的诱变效应,以冬小麦品种新麦18和轮选987的干种子为试验材料,研究了7Li离子束处理对小麦幼苗生长和M1代基因组DNA变异的影响.结果表明,7Li离子束处理虽然对小麦干种子的发芽率和幼苗根的生长无显著影响,但对幼苗高度影响极显著,而且20 Gy剂量的7Li离子束就可使幼苗高降低到对照的一半左右.SSR分析表明,7Li离子束可明显诱发小麦M1代基因组DNA SSR带型的变化.两个品种各处理间的平均多态性频率都达到了13.1%,且多态性片断的类型以扩增片断的增加和缺失为主.以上结果表明,7Li离子束处理小麦种子具有较高的生物损伤效应,可以造成DNA的变异,从而有可能产生可遗传的变异.

Abstract:Dry seeds of two wheat cultivars Xinmai18 and Lunxuan987 were implanted by different doses of ^7Li ion beam to study the effects on seedling growth and DNA variation in M1 generation. The results showed that ^7Li ion beam implantation into wheat seeds can significantly affect seedling height, while no significant effect on germination rate and root growth. The 20 Gy dose of ^7Li ion beam implantation could make the seedling height about 50% shorter than the control. The bands of SSR were greatly changed and the average polymorphic frequency was both 13.1 % in two wheat eultivars implanted by different doses of ^7Li ion beam. The polymorphic patterns were mainly the increase or deletion of amplified fragments. It was concluded that high biological damage effects and DNA variations could be induced by ^7Li ion beam implantation, which could result in the hereditable variation in the later generation.
