性成熟前籽鹅PRL及PRLR mRNA表达量的动态变化

摘    要:选取1日龄、1、2、3、4和5月龄籽鹅各6只,取其垂体和卵巢,利用real-time PCR技术对垂体组织中PRL和卵巢中的PRLR进行实时定量,来探讨PRL及其受体mRNA表达量的变化规律,进而了解卵巢对PRL的反应能力。结果表明籽鹅垂体组织在出雏当日就具有表达PRL的能力,并且表达量较高,从1~3月龄PRL的表达量持续下降,4月龄时开始上调,1~5月龄相对表达量均显著低于1日龄(P0.05),1、5月龄与2~4月龄差异显著(P0.05)。卵巢中PRLR表达量与垂体中PRL表达趋势相近,卵巢PRLR的表达量在1月龄时最高,与其它各月龄之间差异显著(P0.05)。由此可以看出在性成熟之前,PRL及其受体基因的表达均处于较低水平,卵巢对PRL的反应能力较弱,PRL在卵巢发育过程中所起的作用是不明显的。

The Dynamic Changes of PRL and PRLR mRNA of Zi Goose before Sexual Maturity

Abstract:The present experiments were conducted to evaluate the expression profile of PRL and PRL receptor mRNA in the ovary and pituitary of Zi geese before sexual maturity. The expression levels of PRL and PRL receptor mRNA in the ovary and pituitary of Zi geese at the age of 1 day, 1, 2, 3, 4 and 5 month (n=6, respectively) were examined by quantitative real-time (qRT-PCR). The results showed that PRL mRNA expression in the Zi geese pituitary was great at the age of 1 day. The expression in the Zi geese pituitary was greater at the age of 1 to 3 month compared with that at 1 day kept on decreasing , and began to heighten at the age of 4 month. The expressions of PRL mRNA at the age of 1, 2, 3, 4 and 5 month were lower than that at the age of 1 day (P〈0.05), The expressions of PRL mRNA at the age of 1 and 5 month were significant difference compared with that at the age of 2, 3 and 5 month (P〈0.05). The expression of PRLR was close to that of PRL mRNA, and PRLR mRNA expression in the Zi geese ovary was great at the age of 1 day and was higher than that at the age of 1, 2, 3, 4 and 5 month (P〈0.05). It was concluded that the expression of PRL and PRL receptor mRNA in the ovary and appendices suprasphenoidalis of Zi geese before sexual maturity remained at a low level; the reaction ability of ovaries to PRL was weak, and PRL played an unimportant role in the stage of development process of ovaries.
