番茄OSCA基因家族鉴定及不同胁迫条件下表达分析

摘    要:为阐明高渗性钙离子通道OSCA基因在不同胁迫条件下作用,基于番茄全基因组信息,从栽培番茄中鉴定出12个SlOSCA基因,均含DUF221结构域,分别位于1、2、4、6、7、8、9、12号染色体上,亚细胞定位分析表明均位于质膜和高尔基体上。系统进化分析显示SlOSCA基因家族可分为5个进化群。根据番茄表达量数据库组织特异性分析发现,SlOSCA基因家族在不同组织部位表达量不同,其中根部和叶片表达量相对较高。Real-time PCR结果表明,多数SlOSCA基因响应干旱、盐、低温、ABA和灰霉病胁迫。SlOSCA基因响应ABA胁迫普遍强烈,受低温胁迫影响较小。其中SlOSCA9受干旱、盐和ABA胁迫强烈诱导,SlOSCA10受灰霉病胁迫强烈诱导。

Identification of tomato OSCA gene family and expression analysis under different stress conditions

Abstract:To clarify the function and role of calcium channels which were OSCA genes under different stresses, this study identified 12 SlOSCA genes from tomato based on tomatoes genome-wide information, all the genes contained DUF221 domain, tomato SlOSCA genes located on chromosomes 1, 2, 4, 6, 7, 8, 9 and 12, respectively, subcellular localization analysis indicated that they were located on the plasma membrane and golgi. System evolution analysis analysis showed that the SlOSCA genes could be divided into five evolutionary groups. According to the tissue specificity analysis of tomato expression database, the expression levels of SlOSCA gene family in different tissue parts were different, and the expression levels in roots and leaves were relatively high. Real-time PCR results indicated that the great mass of SlOSCA genes could responsive to drought, salt, cold, ABA and gray mold stresses. SlOSCA genes response to ABA stress was generally strong, and its response to cold stress was little. Among them, SlOSCA9 was greatly induced by drought, salt and ABA stress, and SlOSCA10 were greatly induced by gray mold stress.
