摘    要:试验以亚麻、大麻和青麻细胞壁加厚的韧皮纤维为材料,进行细胞骨架制备条件优化研究。即在普通考马斯亮蓝R250染色基础之上,对1%Triton X-100抽提非骨架蛋白时间进行了优化。结果表明,在37℃恒温箱中用1%Triton X-100处理材料30 min,纤维细胞骨架清晰可见。试验为进一步研究细胞骨架系统和纤维素微纤丝如何在细胞壁中沉积等奠定了基础。

Abstract:The bast fibres with the thicken cell wall of flax, hemp and abutilon were used as test materials to study the cytoskeleton morphology, and to optimize the time of non-matrix proteins by 1% Triton X-100 with coomassie brilliant blue R250 staining. The results showed that cytoskeleton of the fibre cell was clearly visible through 30 min treatment by 1% Triton X-100 at 37℃. This experiment established the foundation for studying cytoskeleton system and how the cellulose microfibril deposied in the cell wall in the future.
