摘 要: | 从东北地区的自然环境中初步筛选得到多株纤维素酶高产菌,经过滤纸糖化能力(FPA)、羧甲基纤维素钠酶活力(CMCase)和滤纸崩解能力及失重率测定,获得1株菌T-6,CMCase为65 U、FPA为82.4 U,在培养的第7天滤纸崩解成糊状、失重率为50%。形态、生理生化鉴定和16Sr DNA基因序列鉴定该菌株为缺陷短波单孢菌(Brevundimonas diminuta)。该菌最佳产酶条件:玉米粉0.2%,大豆粉0.5%,K_2HPO_40.6%,Tween-80 0.03%,pH 7.0,培养温度30℃,接种量1%,优化后CMCase达76 U、FPA达92 U。

Isolation and Identification of a Cellulose-Degrading Strain T-6 and Characterization of Its Enzyme Production |

Abstract: | Many strains with high cellulase activity were screened from the natural environment of Ji-lin region. A T-6 strain with the highest cellulase activity was obtained through methods of FPA and CMCase, FPA is as high as 82. 4 U and CMCase is as high as 65 U. Filter paper became paste on the seventh day of culture and weight loss rate is 50%. The strain T-6 was classified as Brevundi-monas diminuta by the analysis of its morphology, physiology and biochemistry, and 16SrDNA gene sequences. The optimal enzyme production conditions are corn flour 0. 2%, soybean meal 0. 5%, K2HPO4 0. 6%, Tween-80 0. 03%, pH 7. 0, culture temperature 30 ℃, inoculation volume 1%. After optimization, FPA is as high as 92 U and CMCase is as high as 76 U. |
