摘    要:从原核表达载体pETChi上切下几丁质酶基因片段,取代植物表达载体pBIUA上的Res基因片段,构建玉米ubi启动子驱动的单子叶高效植物表达载体pBIUC;以目前国内大面积种植的6个骨干自交系综3、87-1、郑58、昌7-2、137、和K12的幼胚为材料,诱导愈伤组织,并以愈伤组织诱导率较高的综3、郑58和K12建立玉米植株再生体系.结果表明:pBIUC植物表达载体成功构建;玉米愈伤组织的诱导对基因型依赖较强;愈伤组织诱导的最佳2,4-D浓度为2.0 mg/L.

Abstract:By replacing fragement Res in plasmid pBIUA with the aimed fragement Chi,cut off from plasmid pETChi,the monocotyledonous higheffective plant expression vector driven by ubi promoter of maize was constructed.Using immature embryos of six elite maize inbred-lines,Zong3,87-1,Zheng58,Chang7-2,137,K12,which were widely used in hybridization,as explants to induce callus.Then establishing regeneration systems of Zong3,Zheng 58 and K12,which had the higher induced frequency of callus.The results showed that the plant expression vector pBIUC were constructed successfully;the callus initiation of maize relies more on genotype;the optimum concentration of 2,4-D induced the callus initiation was(2.0 mg/L).
