香烟烟雾水溶液诱导大蒜根尖细胞产生微核

摘    要:为了探究香烟烟雾水溶液对大蒜根尖细胞遗传物质的影响,通过常规的微核技术观察了香烟烟雾水溶液不同浓度和不同时间处理下根尖细胞中微核的形态和数量。结果表明:在0.5~8支/m L的浓度范围内,大蒜根尖细胞的微核率随着浓度的增加而不断上升,差异极显著(P0.01);在12~36 h处理时间范围内,微核率随着处理时间的增长呈现先升高后降低的趋势;当处理时间为12和24 h时,微核率随着浓度的增加而明显上升,而当处理时间为36 h时,微核率随着浓度的增加呈现先升后降的趋势;香烟烟雾水溶液的浓度和处理时间与微核的产生显著正相关,烟雾浓度对大蒜根尖细胞微核产生的贡献更大;试验组的细胞微核率均极显著高于空白对照组(P0.01),微核指数均大于3.5,属于重度损伤。上述结果表明,香烟烟雾水溶液在较低的浓度(0.5支/m L)和较少的处理时间(12 h)就对植物遗传物质产生严重损伤。

Abstract:In order to explore the influence of the water-soluble fraction of cigarette smoke on the micronucleus of plant cells, choosing the garlic rooy tip as the experimental material and extracting water-soluble fraction of different concentrations, this study analyses the influence of different concentration and treatment time on the genetic material of cells of root tip of Garlic. The results indicated that micronucleus rate increased with the concentration increases when the range of water-soluble fraction of cigarette smoke is 0.5 to 8. The rate of micronucleus increases first and then decreases at last when treatment continued12-36 h. however, when the treatment continued 12 or 24 hours, the rate of micronucleus increases quickly with the growth of concentration. While the treatment continued 36 hour, the rate of micronucleus increases first and then decreases at last. the concentration and treatment time have an interaction on producing micronucleus, and the concentration of water-soluble fraction of cigarette smoke contributes more to producing micronucleus than the treatment time. All the rate of micronucleus of experimental group exceeds that of control group (P<0.05), and the index of micronucleus is bigger than 3.5, belonging to severe damage. In conclusions, under the quite low concentration (0.5/mL) and less treatment time , the water-soluble fraction of cigarette smoke has severe damage to the genetic material of plant.
