摘 要: | [目的]为莲藕种藕的快速繁殖提供参考。[方法]以湖州主栽莲藕品种"迟来早"的地下茎尖为外植体,考察不同消毒时间和消毒方式(0.1%HgCl2、1.0%NaClO+0.1%HgCl2)对外植体的消毒效果,并研究取材时间和培养基中6-BA、NAA浓度对外植体分化的影响。[结果]HgCl2和NaClO联合对外植体消毒效果最好;最佳诱芽培养基为MS+1.0 mg/L6-BA+0.2 mg/L NAA;莲藕地下茎处于冬季休眠期时取材最佳,外植体分化率最高;将再生芽接种到培养基MS+1.0 mg/L6-BA上进行增殖,然后挑选生长良好的芽苗接种到生根培养基MS+0.1 mg/L6-BA+1.0 mg/LIBA+0.2%AC上可正常生根。[结论]该研究确定了莲藕品种"迟来早"地下茎尖的最佳离体培养条件。

Abstract: | [Objective]The study aimed to provide reference for rapid propagation of seed lotus.[Method]With subterraneous stem apex of mainly cultivated lotus variety "Chilaizao" of Huzhou as explant,the sterilization effects of different sterilization time and methods(0.1%HgCl2 and 1.0%NaClO+0.1%HgCl2) on explants were investigated and the influence of sampling time and 6-BA and NAA concn.in medium on the differentiation of explants was studied.[Result]The sterilization effect of combination of HgCl2 and NaClO on explants was best.The optimum adventitious bud-induction medium was MS + 1.0 mg/L 6-BA + 0.2 mg/L NAA.The dormancy stage of subterraneous stem of lotus was optimum time for sampling and its differentiation rate of explants was highest.The regeneration buds were inoculated on the medium MS+1.0 mg/L 6-BA for multiplication,and then the bud seedlings with good growth potential were selected and inoculated on rooting medium MS+0.1 mg/L 6-BA+1.0 mg/L IBA +0.2%AC and they could root normally.[Conclusion] The optimum isolated culture conditions of subterraneous stem apex of lotus variety "Chilaizao" were confirmed in this study. |
