文冠果离体培养技术研究

Abstract:Experiments of in vitro culture were conducted under different hormone combination conditions with MS as basic medium and stems with buds of Xanthoceras sorbifolia as explants.The hormone combinations of MS+6-BA1.00 mg·L-1+NAA0.20 mg·L-1 and MS+6-BA1.00 mg·L-1+NAA 0.50 mg·L-1 of were more suitable for induction(90.4% and 89.3%,respectively) and proliferation(4.5 and 4.8,respectively) of X.sorbifolia and the seedling heights were 1.96 cm and 1.97 cm,respectively.The 1/2MS+IBA0.50 mg·L-1 was the optimal rooting medium,with 93.1% rooting rate,4.8 roots and 1.5 cm root length on average.The temperature and humidity should be restrictedly controlled when seedlings were transplanted.Considering the cost and survival rate,the optimal medium for transplantation was garden soilfurnace ash at 21.After transplanted 30 d,the average height of seedlings was 4.5 cm with above 90% survival rate.
