摘    要:根据基因库中已登录的猪附红细胞体新的基因组DNA序列设计引物,从疑似猪附红细胞体(Mycoplasma suis)感染猪全血样品基因组DNA中扩增出了预期长度666 bp的目的DNA片段,通过对24份临床疑似病例样品的PCR扩增及其他病原微生物基因组DNA的特异性扩增试验,建立了E.suis的PCR诊断方法;进一步对PCR产物克隆、测序分析表明,与国外已报道的AJ504999株相关区域核苷酸、氨基酸序列同源性分别为98.19%和96.85%,表明我国分离株与国外株基因组存在差异。

Abstract:A specific PCR method was developed using the primers designed basing on the novel published genome DNA sequences of Eperythrozoon suis to identify the suspectable E.suis infected pigs.An expected PCR product about 666 bp in length was amplified from blood samples of the symptomatic pigs with clinical signs.The PCR specificity was further evaluated by amplifying genomic DNA from the other pathogenic microorganisms and 24 blood samples taken from suspicious infected pigs basing on the clinical signs and microscope checking.The PCR product was subsequently sequenced,and the homology ratios of nucleotide and amino acid sequences of the PCR product with the corresponding region of published E.suis(AJ504999) genome were 98.19% and 96.85%,respectively,which suggested that the Chinese E.suis strain have varied in genomic DNA compared with the foreign strain.
