陆地棉分子遗传图谱的构建

摘    要:利用SSR引物对具有抗黄萎病性状的F2群体进行分子遗传图谱构建研究.结果表明:2 000对SSR引物在对新陆中10号和军棉1号的多态性筛选中,共筛选到73个稳定的SSR多态性位点.对73个多态性位点在F2群体中进行了验证,χ2 测验表明有6标记位点明显偏分离,67个标记符合χ2 测验,其中共显性标记为61个,显性标记6个;利用Mapmaker/EXP(version 3.0 b)对67个标记构建了连锁群,其中47个标记位点被分配到14个不同的连锁群上,20个标记位点没有分配到连锁群上;14个连锁群总的长度542.7 cM,覆盖棉花基因组的12.2;.首次N1211标记定位在A01染色体上,将N1187标记定位在D08染色体上.

Abstract:In the present paper,upland cotton(Gossypium hirsutum L.) line,Xinluzhong10 with high resistance to Verticillium wilt properties was used as a parent to construct molecular linkage map for identifying molecular markers linked with resistance to Verticillium wilt properties.The stable QTLs affecting resistance to Verticillium wilt traits across different generations will be greatly helpful to be used effectively in molecular marker-assisted selection to improve resistance to Verticillium wilt of cotton cultivars in the future.A total of 2 000 pairs of SSR primers developed were used for the parental survey.There were 73 pairs of SSR primers with DNA polymorphisms between the two parents.After survey and analysis of individuals of 174 plants of the(Xinluzhong10×Junmian1) F_2 using SSR markers,χ~2/χ_c~2-test in F_2 populations showed that 6 primers were significant segregation distortion and 67 SSR markers identified produced a normal segregation of 3∶1 or 1∶2∶1.61 SSR markers were co-dominance and 6 SSR markers were co-dominance.The genetic map for this populations generated 14 linkage groups by Mapmaker/Exp 3.0 included 47 loci covering 542.7 cM,or covering approximtely 12.2% of the total cotton genome.N1211 has been located on A01 of cotton Chromosome and N1187 located on D08 at first.
