人工老化芥菜种子基因组DNA的ISSR标记

摘    要:应用ISSR分子标记对人工老化的芥菜种子进行DNA损伤程度的分析,从18条ISSR引物中选出6条适用于老化芥菜种子的研究分析,并发现人工老化处理的芥菜种子随着老化时间的延长,ISSR扩增出的条带减少、颜色变浅甚至消失,表明在种子贮藏中,随着老化程度的加剧,DNA破损是影响种质资源保存的因素之一。

Abstract:ISSR molecular marker was applied to analyze the DNA damaging degree of artificial aging muster seed. Six primers suitable for studying aging mustard(Brassica juncea Coss.)seeds were selected from 18 primers. We found that along with the prolongation of aging time of muster seed with artificial aging treatment, the numbers of ISSR amplified bands were decreased and their color became light and even faded away. This indicated that during the seed storage, along with the intensification of aging, DNA damages was one of the factors influencing conservation of germplasm resources.
